Combined Reactor and Microelectrode Measurements in Laboratory Grown Biofilms
A combined biofilm reactor-/microelectrode experimental set-up has been constructed, allowing for simultaneous reactor mass balances and measurements of concentration profiles within the biofilm. The system consists of an annular biofilm reactor equipped with an oxygen microelectrode. Experiments were carried out with aerobic glucose and starch degrading biofilms.
The well described aerobic glucose degradation biofilm system was used to test the combined reactor set-up. Results predicted from known biofilm kinetics were obtained. In the starch degrading biofilm, basic assumptions were tested with the microelectrode measurements. It was established, that even with a high molecular weight, non-diffusible substrate, degradation took place in the depths of the biofilm.
Intrinsic enzymatic hydrolysis was not limiting and the volumetric removal rate of oxygen was zero order.
